Optically-controlled Selective Transfection of Partile Sensor Using Multilayered Liposome Containing Photochromic Chemical into a Cell Nucleus

We developed selective transfection of single particle sensor into a specific cell by optically controlled cell immobilization and membrane fusion of liposome for intracellular measurement. The particle was encapsulated in the liposome using packaging based on multi-layering method. The liposomes contain 1,3,3-Trimethylindolino-6'nitrobenzopyrylospiran (SP) that controls the surface potential of the liposome. While the surface potential of the liposome was negative under VIS illumination, it becomes positive by UV-induced photoisolation of SP. The positivelycharged liposome immobilized to the cell membrane by Coulomb attraction. We demonstrated the selective immobilization of the liposome to cells and observed the transfected sensor in cell nucleus.
